PAM resin is one of the most widely used resins in Boc chemical peptide synthesis. Compared to Merrifield resin, it exhibits higher acid stability and is suitable for synthesizing medium-sized and larger peptides, such as DNA-binding polyamines. It is typically coupled to the resin using DCC, HOBT, and DMAP. The remaining part of the peptide can be prepared using standard coupling conditions. The completed peptide can be simultaneously deprotected and cleaved from the resin using hydrofluoric acid. Hydrofluoric acid is highly toxic, volatile, and corrosive, and must be used in special hydrofluoric acid-resistant equipment.
